In the present experiment, we showed that 3-amino-1,2,4-triazole (3-AT) inhibits seed germination of the loss-of-function mutant abi5-1, but promotes seed germination of the ABI5-overexpression transgenic line, and that ABI5 affects reactive oxygen species (ROS) homeostasis. ABI5 overexpression rescued the growth arrest phenotype at low CO 2. Taken together, our results demonstrate that PPC2 and ABI5 are key regulators of plant acclimation to low CO 2, and positively contribute to carbon fixation and metabolism in C 3 plants. Pharmacological assay showed that abi5-1 mutant was insensitive to TOR inhibitor AZD8055, whereas AtABI5 overexpression lines were hypersensitive to AZD8055 in Arabidopsis. Biochemical interaction assays demonstrated that ABI5 physically interacted with the RIBOSOMAL S6 KINASE2 (S6K2) protein in plant cell.
